『マスエフェクト3』は、シェパード司令官が、地球に侵攻し、銀河系の文明を次々と征服している機械の種族「リーパー」に対して、最後の戦いを繰り広げるアクションRPGです。 プレイヤーは様々な種族やクラスから分隊メンバーを募集し、同盟を結び、どのキャラクターが生き残るかを決定する選択を行います。戦闘では、バイオティック能力とテック能力を組み合わせた分隊ベースの戦術、ブラインドファイア、強化された近接攻撃が特徴です。
本作には、最大4人のプレイヤーが協力して敵の波状攻撃を撃退し、紛争地域を解放する協力プレイモードが搭載されています。マルチプレイヤーでの進捗は、シングルプレイヤー・キャンペーンにおける戦備の強化につながります。兵士のジェームズ・ベガや、実体を持ったAIであるEDIなど、新規およびお馴染みのチームメンバーがシェパードのチームに加わります。
