『Destroy All Humans!』シリーズ第1作目となるアクションアドベンチャーゲーム『Destroy All Humans!』のリメイク版。舞台は1959年のアメリカ。本作は、当時のライフスタイル、ポップカルチャー、政治的風潮を風刺している。 プレイヤーは、「フロン」と呼ばれる架空の異星人種族の一員であるクリプトスポリジウム137となり、フロン族のスパイ兼暗殺者として、自種族のクローン作成プロセスを維持するために人間のDNAを採取するという使命を帯びて地球に侵入する。
このリメイク版では、新たなミッション、操作体系、アップグレード、スキン、能力、そしてグラフィックの改良が盛り込まれている。
